Effects of acetic acid, ethanol and SO2 on the removal of volatile acidity from...

Moura, A. Vilela; Schuller, Dorit Elisabeth; Faia, A. Mendes; Côrte-Real, Manuela

Herein we report the influence of different combinations of initial concentration of acetic acid and ethanol on the removal of acetic acid from acidic wines by two commercial Saccharomyces cerevisiae strains S26 and S29. Both strains reduced the volatile acidity of an acidic wine (1.0 g l-1 acetic acid and 11% (v/v) ethanol) by 78% and 48%, respectively. Acetic acid removal by both strains was associated with a...

Effect of refermentation conditions and microoxygenation on the reduction of vo...

Moura, A. Vilela; Schuller, Dorit Elisabeth; Faia, A. Mendes; Côrte-Real, Manuela; Falco, V.

Herein, we evaluate the applicability of previously characterized commercial and indigenous Saccharomyces cerevisiae strains and non-S. cerevisiae species for the deacidification of white and red wines at a pilot scale. The effect of the refermentation process (mixture of acidic wine with musts from freshly crushed grapes or with residual marc) as well as MO (MO) on acetic acid removal efficiency and wine aroma...

Amplification of genomic sequences flanked by delta elements of retrotransposons TY1 and TY2 is a reliable method for characterization of Saccharomyces cerevisiae strains. The aim of this study is to evaluate the usefulness of microfluidic electrophoresis (Caliper LabChip®) to assess the factors that affect interlaboratory reproducibility of interdelta sequence typing for S. cerevisiae strain delimitation. We c...

A new high-throughput method for the rapid extraction of yeast DNA

Vieira, E.; Neves, J. Drumonde; Lima, Maria Teresa; Araújo, Isabel M.; Casal, Margarida; Schuller, Dorit Elisabeth

Ecological, evolutionary and population genetic studies of yeasts require the processing of a high numbers of isolates. The current molecular methods for yeast identification and characterization require time-consuming and labor intensive DNA extraction protocols. Within this research, we aimed to develop a high-throughput method for yeast DNA extraction. Total yeast DNA extraction was based on a previously des...

Schuller, Dorit Elisabeth; Cardoso, Filipa; Sousa, Susana; Gomes, Paula; Gomes, Ana Catarina; Santos, Manuel A. S.; Casal, Margarida

We herein evaluate intraspecific genetic diversity of fermentative vineyard-associated S. cerevisiae strains and evaluate relationships between grape varieties and geographical location on populational structures. From the musts obtained from 288 grape samples, collected from two wine regions (16 vineyards, nine grape varieties), 94 spontaneous fermentations were concluded and 2820 yeast isolates were obtained ...

Yeast biodiversity in vineyard environments is increased by human intervention

Drumonde-Neves, Joao; Franco-Duarte, Ricardo; Lima, Teresa; Schuller, Dorit; Pais, Célia

One hundred and five grape samples were collected during two consecutive years from 33 locations on seven oceanic islands of the Azores Archipelago. Grape samples were obtained from vineyards that were either abandoned or under regular cultivation involving common viticultural interventions, to evaluate the impact of regular human intervention on grape yeast biota diversity in vineyards. A total of 3150 yeast i...

Association between Grape Yeast Communities and the Vineyard Ecosystems

Drumonde-Neves, Joao; Franco-Duarte, Ricardo; Lima, Teresa; Schuller, Dorit; Pais, Célia

The grape yeast biota from several wine-producing areas, with distinct soil types and grapevine training systems, was assessed on five islands of Azores Archipelago, and differences in yeast communities composition associated with the geographic origin of the grapes were explored. Fifty-seven grape samples belonging to the Vitis vinifera grapevine cultivars Verdelho dos Acores (Verdelho), Arinto da Terceira (Ar...
